Approaches to the Risk Assessment of Genotoxic Carcinogens in Food: A Critical Appraisal ANNEX Introduction to Genetic Toxicology
نویسندگان
چکیده
This review document addresses the processes of DNA damage, repair and mutation as key steps on the pathway towards cancer development and also briefly describes the most common genotoxicity tests used in the evaluation of whether a compound is genotoxic or not. Knowledge that the carcinogen under evaluation is genotoxic or nongenotoxic is a vital stage of the risk assessment and influences the handling of the human health issues arising from the compound. It is not possible within the scope of this document to provide guidance on the strategies used for concluding whether a compound is genotoxic as this can be a very complex evaluation and is best conducted with expert input. However it is hoped that this review provides an insight into the background behind genotoxicity testing, leading to a better understanding of the genotoxicity tests involved, what they can and cannot predict and the underlying processes that lead to the data produced.

Approaches to the risk assessment of genotoxic carcinogens in food: a critical appraisal.
The present paper examines the particular difficulties presented by low levels of food-borne DNA-reactive genotoxic carcinogens, some of which may be difficult to eliminate completely from the diet, and proposes a structured approach for the evaluation of such compounds. While the ALARA approach is widely applicable to all substances in food that are both carcinogenic and genotoxic, it does not...

This report compares cancer classification systems, health risk assessment approaches, and procedures used for establishing occupational exposure limits (OELs), in various European countries and scientific organizations. The objectives were to highlight and compare key aspects of these processes and to identify the basis for differences in cancer classifications and OELs between various scienti...

This paper presents the work of an expert group established by the International Life Sciences Institute - European branch (ILSI Europe) to follow up the recommendations of an international conference on "Risk Assessment of Compounds that are both Genotoxic and Carcinogenic: New Approaches". Twelve genotoxic and carcinogenic chemicals that can be present in food were selected for calculation of...
متن کاملAppraisal of the evolutionary-based methodologies in generation of artificial earthquake time histories
Through the last three decades different seismological and engineering approaches for the generation of artificial earthquakes have been proposed. Selection of an appropriate method for the generation of applicable artificial earthquake accelerograms (AEAs) has been a challenging subject in the time history analysis of the structures in the case of the absence of sufficient recorded accelerogra...
